Overview

ONT-10 is a therapeutic vaccine designed to direct an individual's immune system to identify and destroy cancer cells. ONT-10 is a liposomal formulated vaccine composed of a 43 amino acid glyocoprotein sequence of the tumor associated antigen MUC-1 and the novel, synthetic adjuvant, PET-Lipid A, which is proprietary to Oncothyreon.

ONT-10 is designed to produce both an antibody and a T-cell immune response to cancer cells that express the MUC-1 target. IND enabling studies of ONT-10 are currently underway at Oncothyreon. Oncothyreon currently expects to filed an Investigational New Drug application for ONT-10 in December 2011 (View press release) fourth quarter of 2011 and plans to begin a Phase 1 clinical trial by the first half quarter of 2012.

Preclinical results on ONT-10 were presented at the American Association for Cancer Research Conference in 2011 ( View press release | View poster). Results demonstrated that administration of ONT-10 produced a robust antibody response in mice that was specific for human tumor MUC-1. A strong cellular immune response directed to the target was also shown. ONT-10 blocked the growth of two different tumors expressing human MUC-1 in murine models, with a high proportion of animals tumor free at the conclusion of the experiment. Additionally, the adjuvant component of ONT-10, PET-Lipid A, a fully synthetic toll like receptor 4 (TLR4) agonist developed by Oncothyreon, was shown to have enhanced potency compared to the adjuvant monophosphoryl lipid A (MPL).
